Highlights

In this application note, we have demonstrated a workflow to create highly reproducible and consistent advanced liver models in 3D with RASTRUM™. The advanced liver cell model was treated with TGF-β1 into a profibrogenic phenotype and fibrosis impact was quantified by several downstream analyses.

  • Easily co-culture of PH5CH8 human hepatocytes and LX-2 human stellate cells in RASTRUM™ Matrix to maintain a more native state of both cells.
  • Primary hepatocytes cultured prepared using RASTRUM™ Platform remains viable after 14 days of culture.
  • Liver model can be treated with TGF-beta to trigger fibrosis successfully.
  • RASTRUM™ Liver Fibrosis model validated using qPCR showing an increase in COL1A1 and CTGF signal compared to non-treated samples.
